An Integrated Microfluidic System for Sample Concentration and Extraction

Many patients who are infected with HIV but are on treatment have low viral loads which cannot be detected with conventional qRT-PCRs. The integrated system we have designed concentrates a human plasma sample with defective HIV to make it more easily detectable using less expensive diagnostic techniques. The viral RNA is then extracted using a low-cost solid phase extraction column following virion lysis. qRT-PCR can then be used to measure the viral load from the extracted RNA. This concentration platform improves the limit of detection of conventional qRT-PCR and can be applied in resource limited settings due to its low cost components. This point-of-care concentration device is a step towards a more accessible HIV diagnostic platform.
